This is a working overview of tuftsin, written for readers who want more than a one-paragraph summary but less than a textbook.
This page was last updated on 2025-08-14 and is reviewed periodically as new material appears.
Enzymatic stability motivates the extra three residues at the carboxyl end. Native tuftsin is cleaved quickly by circulating aminopeptidases and carboxypeptidases, which limits its duration of action and its usefulness as a tool compound. Extending the chain with proline-rich segments is a common design tactic because proline constrains the backbone and slows proteolysis. The same Pro-Gly-Pro motif appears in other Russian-developed peptides of the era. Whether the full seven-residue chain is required for activity, or whether it acts mainly as a prodrug releasing tuftsin, remains unresolved.
Selank is a synthetic heptapeptide with the sequence Thr-Lys-Pro-Arg-Pro-Gly-Pro. Its four N-terminal residues reproduce tuftsin, a tetrapeptide fragment of the immunoglobulin heavy chain, while the C-terminal Pro-Gly-Pro extension is a synthetic addition. The peptide has a molecular mass near 752 daltons and carries a net positive charge at physiological pH because of the arginine and lysine side chains. Published indexes list it under the name Selank and the sequence abbreviation TKPRPGP. Solid-phase peptide synthesis is the standard production route for research quantities.
Handling follows standard practice for research peptides. Material is weighed in a low-humidity environment because the powder absorbs atmospheric moisture. Purity is reported as the percentage area of the main peak in a chromatogram, with specifications commonly set at 95 percent or higher; values below that threshold indicate the presence of truncated or modified species. Residual trifluoroacetate from purification is often present and may affect mass balance. Certificates of analysis should state the analytical method, the column and gradient used, and the lot-specific retention time so that results can be compared across suppliers.
Identity and purity of selank are established with reversed-phase high-performance liquid chromatography coupled to mass spectrometry. The peptide elutes from C18 columns with acetonitrile gradients in water containing trifluoroacetic acid or formic acid, and detection is usually performed by ultraviolet absorbance near 214 nm. Electrospray ionization in positive mode gives a doubly protonated ion near m/z 377, consistent with a mass of about 752 Da. Amino acid analysis or tandem mass spectrometry of fragment ions confirms the sequence. Because the molecule has no aromatic residues, it lacks a usable 280 nm chromophore, so low-wavelength detection or mass spectrometry is required.
Peptide bonds in selank are susceptible to hydrolysis under strongly acidic or basic conditions, and the terminal proline residues are vulnerable to exopeptidase activity in biological samples. Lyophilized powder stored dry at -20 °C typically remains stable for extended periods, whereas aqueous solutions degrade faster and may lose measurable purity within days to weeks depending on pH, temperature, and microbial load. Repeated freeze-thaw cycles promote aggregation and adsorption to container surfaces. For analytical work, solutions are usually prepared fresh, kept cold, and used within a single working day.
| Property | Value | Notes |
|---|---|---|
| Chemical class | Synthetic heptapeptide | Tuftsin analogue with a C-terminal Pro-Gly-Pro tail |
| Amino acid sequence | Thr-Lys-Pro-Arg-Pro-Gly-Pro | Abbreviated TKPRPGP in most catalogues |
| Molecular mass | About 752 Da | Value for the free peptide |
| Solubility | Freely soluble in water | Aqueous solutions are kept cold and used promptly |
| Typical storage | -20 °C, dry, protected from light | Applies to the lyophilised powder before reconstitution |
Once dissolved, the peptide is markedly less stable than the dry powder. Aqueous solutions are subject to backbone hydrolysis and to microbial growth when they are handled without sterile technique. Buffered solutions near neutral pH are common for short-term laboratory work, while acidic conditions are sometimes used to improve solubility. Analytical laboratories generally prepare working solutions fresh rather than storing them, and a residual water film left in a reopened vial can seed degradation even when the container appears dry.
Reversed-phase high-performance liquid chromatography is the standard technique for estimating peptide purity. The result is a peak-area percentage, which describes how much of the detected material elutes as the main peak in one run. Mass spectrometry confirms the molecular mass and can reveal truncated, adducted, or otherwise modified species. Amino acid analysis or tandem mass spectrometry can address sequence fidelity when identity is in doubt. None of these measurements, taken alone, establishes that a sample is fit for any specific purpose.
Lyophilized selank is normally supplied as a dry powder and is considered stable for extended periods when kept cold and dry. Moisture uptake is the main practical threat, because absorbed water promotes both hydrolysis and aggregation in the solid state. Vials are usually warmed to room temperature before opening so that condensation does not form on the powder. Supplier documentation commonly specifies -20 °C for routine storage, with -80 °C used for material intended to be archived for years.
Selank is a synthetic heptapeptide developed in Russia. Its sequence is Thr-Lys-Pro-Arg-Pro-Gly-Pro, a seven-residue chain built around the natural tetrapeptide tuftsin. Researchers at the Institute of Molecular Genetics of the Russian Academy of Sciences first described the compound in the mid-1990s. The design combined the tuftsin core with an added Pro-Gly-Pro tail, a modification intended to extend the molecule's stability in biological fluids. Published work on the peptide has appeared mainly in Russian-language journals.
Reported activity for Selank centers on anxiolytic and nootropic effects. Russian clinical reports describe use in anxiety and in cognitive or attention-related complaints. Most of this evidence comes from studies conducted by the same research groups that developed the peptide. Independent replication in other countries remains limited, and no major Western regulatory agency has approved the compound for any indication. The gap between local reports and external verification is a recurring point in discussions of the peptide.
Selank is a synthetic heptapeptide developed in Russia as a structural analogue of tuftsin, a naturally occurring immunomodulatory tetrapeptide. Its sequence, Thr-Lys-Pro-Arg-Pro-Gly-Pro, keeps the tuftsin core at the N-terminus and appends a Pro-Gly-Pro tail. Researchers at the Institute of Molecular Genetics in Moscow synthesized the compound during the 1990s while searching for peptides with combined anxiolytic and immunomodulatory activity. The added tail was intended to resist enzymatic cleavage and prolong the molecule's presence in circulation.
The compound has a calculated molecular weight near 751.9 daltons and carries a net positive charge at physiological pH because of its arginine residue. It dissolves freely in water and in common aqueous buffers, and typically appears as a white or off-white lyophilized powder. The amide backbone makes the molecule susceptible to peptidases, which limits oral use and favors intranasal or parenteral routes. Nomenclature in the literature varies: the substance is also described by the sequence abbreviation TP-7 and by a Russian trade designation.
Regulatory status differs sharply by region. Selank holds a Russian marketing authorization, where it is supplied mainly as nasal drops, while authorities elsewhere have not approved it for medical use. Material sold internationally is therefore usually labeled as a research chemical rather than a medicine. Peer-reviewed publications come predominantly from Russian laboratories, and sample sizes are generally small. Whether the compound produces comparable effects under independent, well-controlled replication remains an open question that the broader literature has not settled.
It was, in fact, nothing like material used by the Romans, but was a "natural cement" made by burning septaria – nodules that are found in certain clay deposits, and that contain both clay minerals and calcium carbonate. The burnt nodules were ground to a fine powder. This product, made into a mortar with sand, set in 5–15 minutes. The success of "Roman cement" led other manufacturers to develop rival products by burning artificial hydraulic lime cements of clay and chalk. Roman cement quickly became popular but was largely replaced by Portland cement in the 1850s.
==== Edema and connective tissue changes ==== As the blood-nerve-barrier breaks down, proteins and cells will be able to enter the perineural and endoneurial space. The increased permeability allowing substances to enter combined with the lack of a lymphatic system to drain fluids causes an increase in pressure and may alter the ionic environment. The increased pressure in the endoneurium can cause a mini "compartment syndrome" leading to post-traumatic ischemia of the nerve cells. When endoneurial edema is triggered, the swelling will last many hours the point that the entrapment is relieved. For example, following 2-8 hours of compression, endoneurial fluid pressure will rapidly rise and can stay elevated for 24 hours. Lymphocytes, fibroblasts, and macrophages will also be able to cross the newly permeable blood nerve barrier and react to the antigens contained in the perineural space triggering an inflammatory reaction. As part of this inflammatory reaction, there will be excess deposits of fibrin (i.e. scarring). On histological analysis, epineural fibrosis and perineural thickening can be seen. This scarring is an irreversible change associated with nerve entrapment. If the scarring damages the microcirculatory environment, then the impaired blood supply will also be permanent. In cases where permanent impairment exists even after a nerve decompression, it's thought that the pathophysiological basis is due to extensive scarring along and within the nerve, as demyelination and axonmetesis are generally capable of healing but scarring cannot be reversed.
=== Other symptoms and comorbidities === Myoglobinuria (reddish-brown urine) may be seen due to the breakdown of skeletal muscle known as rhabdomyolysis (a condition in which muscle cells breakdown, sending their contents into the bloodstream). In 2020, the largest study to-date of 269 GSD-V patients, 39.4% reported no previous episodes of myoglobinuria and 6.8% had normal CK (including those with fixed muscle weakness); so an absence of myoglobinuria and normal CK should not rule out the possibility of the disease. Between 33-51.4% develop fixed muscle weakness, typically of the trunk and upper body, with the onset of muscle weakness usually occurring later in life (40+ years of age). Younger people may display unusual symptoms, such as difficulty chewing, swallowing, or utilizing normal oral motor functions. Idiopathic leg pains were common in children, usually occurring at night, often presumed to be "growing pains" and not investigated further. Several comorbidities were found in GSD-V individuals at a higher rate than in the general population, including (but not limited to): hypertension (17%), endocrine diseases (15.7%), musculoskeletal/rheumatic disease (12.9%), hyperuricemia/gout (11.6%), gastrointestinal diseases (11.2%), neurological disease (10%), respiratory disease (9.5%), and coronary artery disease (8.3%). They may have a pseudoathletic appearance of muscle hypertrophy (24%), particularly of the legs, and may have lower bone mineral content and density in the legs.
==== Other early Americans ==== American officer John Parker Boyd partook in the Battle of Kharda, fighting on the side of the Nizam of Hyderabad. American Founding Father Aaron Burr had a relationship with an East Indian woman named Mary Emmons, who was most likely from the Indian city of Calcutta. Together, they had two children, including John Pierre Burr. Dudley Leavitt Pickman was an early American trader with India who founded the East India Marine Society. Fitzedward Hall was the first American to edit a Sanskrit text.
== Signs and symptoms == The consumption of large quantities of Lathyrus seeds containing high concentrations of the neurotoxic glutamate analogue β-oxalyl-L-α,β-diaminopropionic acid (ODAP, also known as β-N-oxalyl-amino-L-alanine, or BOAA) causes paralysis, characterized by lack of strength in or inability to move the lower limbs, and may involve pyramidal tracts, producing signs of upper motor neuron damage. The toxin may also cause aortic aneurysm. A unique symptom of lathyrism is the atrophy of gluteal (buttocks) muscles. ODAP is a poison of the mitochondria, leading to excess cell death, especially in motor neurons. Children can additionally develop bone deformity and reduced brain development.

Periodization refers to the organization of training into sequential phases and cyclical periods, and the change in training over time. The simplest strength training periodization involves keeping a fixed schedule of sets and reps (e.g. 2 sets of 12 reps of bicep curls every 2 days), and steadily increasing the intensity on a weekly basis. This is conceptually a parallel model, as several exercises are done each day and thus multiple muscles are developed simultaneously. It is also sometimes called linear periodization, but this designation is considered a misnomer. Sequential or block periodization concentrates training into periods ("blocks"). For example, for athletes, performance can be optimized for specific events based on the competition schedule. An annual training plan may be divided hierarchically into several levels, from training phases down to individual sessions. Traditional periodization can be viewed as repeating one weekly block over and over. Block periodization has the advantage of focusing on specific motor abilities and muscle groups. Because only a few abilities are worked on at a time, the effects of fatigue are minimized. With careful goal selection and ordering, there may be synergistic effects. A traditional block consists of high-volume, low-intensity exercises, transitioning to low-volume, high-intensity exercises. However, to maximize progress to specific goals, individual programs may require different manipulations, such as decreasing the intensity and increasing volume.
=== Other uses and similar symbols === The main attribute of Vishnu is a wheel like weapon called the Sudarśanacakra. Similar wheel symbols were used as a solar symbol by the Ancient Egyptians. Some Buddha statues also depict the related Dharmachakra Mudrā, a hand sign depicting the turning of the Dharma wheel. A very similar wheel symbol also appears in the flag of the Romani people, hinting to their nomadic history. In non-Buddhist cultural contexts, an eight-spoked wheel resembles a traditional ship's wheel. As a nautical emblem, this image is a common sailor tattoo, which may be misidentified as a dharmachakra or vice versa. The sonnenrad is a similar symbol used by occultists and neo-nazis. Falun Gong uses the concept of a similar wheel as a central concept. In the Unicode computer standard, the dharmachakra is called the "Wheel of Dharma" and found in the eight-spoked form. It is represented as U+2638 (☸). As emoji: ☸️.
If SO3 is condensed above 27 °C, then α-SO3 forms, which has a melting point of 62.3 °C. α-SO3 is fibrous in appearance. Structurally, it is the polymer [S(=O)2(μ-O)]n. Each end of the polymer is terminated with OH groups. β-SO3, like the alpha form, is fibrous but of different molecular weight, consisting of a hydroxyl-capped polymer, but melts at 32.5 °C. Both the gamma and the beta forms are metastable, eventually converting to the stable alpha form if left standing for sufficient time. This conversion is caused by traces of water. Relative vapor pressures of solid SO3 are alpha < beta < gamma at identical temperatures, indicative of their relative molecular weights. Liquid sulfur trioxide has a vapor pressure consistent with the gamma form. Thus heating a crystal of α-SO3 to its melting point results in a sudden increase in vapor pressure, which can be forceful enough to shatter a glass vessel in which it is heated. This effect is known as the "alpha explosion".

== Medical uses == As of 2022, across the European countries where etifoxine has been approved (including France, Luxembourg, Malta, Romania and Bulgaria), the main approved indication is for the treatment of the psychosomatic manifestations of anxiety. When etifoxine was first approved in France in 1979, the original indication was for the treatment of "psychosomatic manifestations of anxiety, for instance autonomic dystonia, particularly with cardiovascular expression". Over time, the indication for etifoxine has been more formalized as the treatment of adjustment disorder (situational depression) with anxiety (ADWA) (for example, stress-related anxiety). Etifoxine has been found to reduce scores on the Hamilton Anxiety Rating Scale (HAM‑A) in people with adjustment disorder with anxiety by approximately 50 to 75% after 4 weeks of treatment in clinical trials (as per the AMETIS, ETILOR, ETIZAL, STRETI studies). The medication is similarly effective or more effective than benzodiazepines like lorazepam, alprazolam, and clonazepam and more effective than buspirone for adjustment disorder with anxiety on the basis of directly comparative randomized controlled trials. However, in the AMETIS study, both etifoxine and lorazepam failed to show greater effectiveness over placebo. In the trials comparing etifoxine to clonazepam, lorazepam, and alprazolam, total daily doses of the benzodiazepines were limited to their maintenance daily dose, set at 1 mg clonazepam, 2 mg lorazepam, and 1.5 mg alprazolam, divided across 3 doses per day.
== Treatment == There have been no controlled studies to define the optimal treatment for BPDCN. Studies on small numbers of individuals with the disease have found that the standard chemotherapy regimens used for the initial induction treatments of AML, acute lymphoblastic leukemia, and high-grade lymphoma give complete remission rates of 77%, 93%, and 80%, respectively, in childhood PBDN and 47%, 77%, and 53%, respectively, in adult PBDN. However, these remissions were short-lived: post-treatment mean times to relapse or death were 12 months for children and 6.8 months for adults. Given these poor remission and survival rates, other treatments have been added to the initial treatment regimens. Studies have shown that the addition of intrathecally administered drugs (administered directly into the spinal canal) as prophylaxis prolongs the period of CNS-free disease and increases overall survival. Hematopoietic stem cell transplantation following initial chemotherapy-induced remission also prolongs these remissions and, it is suggested, offers potential for curing the disease. (A graft-versus-leukemia effect may have contributed to the benefits seen after transplantation.) Studies have not yet determined whether allogenic (i.e. taken from others) or autologous (i.e. taken from self) stem cells achieve better results, although one retrospective study in Japan found that autologous stem cells gave significantly better overall and progression-free survival rates.

Selank is a seven-amino-acid peptide built from the tuftsin sequence plus a Pro-Gly-Pro tail. It is produced by chemical synthesis rather than extracted from a natural source. The free peptide is usually supplied as a lyophilised powder or in an aqueous formulation.
Tuftsin is a natural tetrapeptide derived from the Fc portion of immunoglobulin G. Selank retains that tetrapeptide at its N-terminus and adds three residues to improve resistance to enzymatic breakdown. The relationship is structural, and the two molecules are not interchangeable in experimental work.
It holds a product registration in Russia for intranasal use as an anxiolytic. Registrations of that kind are not automatically accepted by regulators elsewhere. In most other countries it is treated as an unapproved substance or a research material.
Identity is confirmed by matching the retention time in reversed-phase chromatography against a reference standard and by measuring the molecular mass with mass spectrometry. Tandem mass spectrometry or amino acid analysis can verify the sequence of the seven residues. Because the peptide contains no aromatic amino acids, detection at 280 nm is not useful.
