=== Names === In 2013, trastuzumab emtansine was approved in the United States with the generic name "ado-trastuzumab emtansine", rather than the original United States Adopted Name (USAN) issued in 2009, "trastuzumab emtansine". Trastuzumab is the anti-HER2 antibody; emtansine refers to the linker-drug (SMCC-DM1). The "ado-" prefix was added at the request of the FDA to help prevent dispensing errors. During preclinical development and clinical trials, the drug was also known as trastuzumab-DM1 or trastuzumab-MCC-DM1 (after the codename for emtansine), both abbreviated T-DM1, and by the codename PRO132365.
=== Environmental pollution === Considering the complex interactions between humans, animals and the environment, it is also important to consider the environmental aspects and contributors to antimicrobial resistance. One of these factors is human and hospital waste, which is a substantial contributor to environmental antimicrobial resistance (AMR). It is estimated that around 50–80% of antibiotics are passed unmetabolized in urine and 4–30% in feces, causing large quantities of active antibiotics to enter sewage systems. Sewage environments contain high levels of antimicrobial-resistant bacteria (ARB), antimicrobial resistance genes (ARGs), and mobile genetic elements such as plasmids, which enable rapid gene exchange. Wastewater treatment plants (WWTPs) are typically not designed to remove AMR; in some cases, multidrug-resistant species such as Escherichia, Shigella, and Klebsiella can increase twofold in treated wastewater. Methicillin-resistant Staphylococcus aureus (MRSA) has also been detected in both raw and treated sewage. Agriculture is also a major contributor to environmental AMR. Manure from livestock (such as cattle, sheep, etc.) contains residual antibiotics, which enter the soil as farmers usually use it as fertilizer. == Technical characteristics == Modern systems (for smaller, i.e. “normal”, tube diameters as used in the transport of small capsules) reach speeds of around 7.5 m (25 ft) per second, though some historical systems already achieved speeds of 10 m (33 ft) per second. RFID chips within the capsules gather data – e. g. about the location of the carrier, its transport speed or its destination. == Addiction == Cocaine addiction results from complex molecular changes in the brain following multiple exposures to cocaine. Dynorphins have been shown to be an important part of this process. Although a single exposure to cocaine does not affect brain dynorphin levels, repeated exposures to the drug increases dynorphin concentrations in the striatum and substantia nigra in rats. One proposed molecular mechanism for increased dynorphin levels involves transcriptional regulation by CREB (3’, 5’-monophosphate response element binding protein). According to the model proposed by Carlezon et al., use of cocaine increases the expression of cAMP and cAMP-dependent protein kinase (PKA). PKA leads to the activation of CREB, which increases the expression of dynorphin in the nucleus accumbens and dorsal striatum, brain areas important in addiction. Dynorphin decreases dopamine release by binding to KORs on dopamine nerve terminals.
=== Pharmacodynamics === MDDMA shows reduced potency as a monoamine releasing agent and reuptake inhibitor compared to MDA and MDMA. It was 11-fold less potent than MDMA and 4-fold less potent than MDA as a serotonin releasing agent (SRA). Moreover, whereas MDA and MDMA are serotonin–norepinephrine–dopamine releasing agents (SNDRAs), MDDMA is a selective SRA along with ≥10-fold weaker dopamine and norepinephrine reuptake inhibition. The related drug MDTMA is completely inactive as a monoamine releasing agent, though it does still show very weak monoamine reuptake inhibition. Another related drug, dimethylamphetamine, is said to be a prodrug of methamphetamine and amphetamine, although it is much less potent and weaker than these drugs. (R)-MDDMA has been found be inactive as a serotonin releasing agent but to act as a partial agonist of the serotonin 5-HT2A and 5-HT2C receptors. Conversely, unlike MDMA, it was inactive as a serotonin 5-HT2B receptor agonist. In animal studies, (R)-MDDMA did not produce the head-twitch response, affect body temperature, or induce hyperlocomotion, and showed diminished or no prosocial effects. On the other hand, it produced psychoplastogenic effects mediated by serotonin 5-HT2 receptor activation as well as promoted fear extinction and induced antidepressant-like effects. It was concluded that (R)-MDDMA is a non-hallucinogenic psychoplastogen with improved safety compared to MDMA and (R)-MDMA.

The use of displacement chromatography is rather limited, and is mostly used for preparative chromatography. The basic principle is based on a molecule with a high affinity for the chromatography matrix (the displacer) which is used to compete effectively for binding sites, and thus displace all molecules with lesser affinities. There are distinct differences between displacement and elution chromatography. In elution mode, substances typically emerge from a column in narrow, Gaussian peaks. Wide separation of peaks, preferably to baseline, is desired in order to achieve maximum purification. The speed at which any component of a mixture travels down the column in elution mode depends on many factors. But for two substances to travel at different speeds, and thereby be resolved, there must be substantial differences in some interaction between the biomolecules and the chromatography matrix. Operating parameters are adjusted to maximize the effect of this difference. In many cases, baseline separation of the peaks can be achieved only with gradient elution and low column loadings. Thus, two drawbacks to elution mode chromatography, especially at the preparative scale, are operational complexity, due to gradient solvent pumping, and low throughput, due to low column loadings. Displacement chromatography has advantages over elution chromatography in that components are resolved into consecutive zones of pure substances rather than "peaks".
==== Aromatase and estrogenicity ==== Testosterone can be metabolized by aromatase into estradiol, and many other AAS can be metabolized into their corresponding estrogenic metabolites as well. As an example, the 17α-alkylated AAS methyltestosterone and metandienone are converted by aromatase into methylestradiol. 4,5α-Dihydrogenated derivatives of testosterone such as DHT cannot be aromatized, whereas 19-nortestosterone derivatives like nandrolone can be but to a greatly reduced extent. Some 19-nortestosterone derivatives, such as dimethandrolone and 11β-MNT, cannot be aromatized due to steric hindrance provided by their 11β-methyl group, whereas the closely related AAS trestolone (7α-methyl-19-nortestosterone), in relation to its lack of an 11β-methyl group, can be aromatized. AAS that are 17α-alkylated (and not also 4,5α-reduced or 19-demethylated) are also aromatized but to a lesser extent than is testosterone. However, it is notable that estrogens that are 17α-substituted (e.g., ethinylestradiol and methylestradiol) are of markedly increased estrogenic potency due to improved metabolic stability, and for this reason, 17α-alkylated AAS can actually have high estrogenicity and comparatively greater estrogenic effects than testosterone. The major effect of estrogenicity is gynecomastia (woman-like breasts).
