This is a working overview of blood-brain barrier, written for readers who want more than a one-paragraph summary but less than a textbook.
This page was last updated on 2026-02-19 and is reviewed periodically as new material appears.
Proposed mechanisms centre on the GABAergic system. Animal and tissue studies report changes in GABA-A receptor expression and reduced activity of GABA transaminase, the enzyme that degrades GABA. Effects on monoamine turnover, including serotonin and dopamine pathways, are also described, and a separate line of work links the peptide to increased expression of brain-derived neurotrophic factor in hippocampal tissue. Most of these findings come from rodent models and cell preparations. How the individual observations combine into a single coherent mode of action is not settled.
Pharmacokinetic data are sparse and largely derived from animal work. After intranasal administration the peptide appears in plasma within minutes, and reported half-lives are short, on the order of minutes to tens of minutes. Degradation proceeds through ordinary proteolytic cleavage into constituent amino acids and smaller fragments. Direct evidence that intact Selank reaches brain tissue in meaningful amounts is limited, and the extent of blood-brain barrier penetration is debated. Some authors argue that fragments, not the parent peptide, carry much of the observed activity.
Published clinical work is concentrated in Russian-language journals and generally involves small samples without independent replication. Systematic reviews in English note the shortage of randomised, placebo-controlled trials and the difficulty of verifying methods from translated reports. Outcome measures vary between studies, which complicates pooling of results. Interest in the compound as a cognitive or anxiolytic agent therefore rests on a thinner evidence base than the volume of citations suggests. Replication in well-powered trials with preregistered endpoints would be needed before firm conclusions about efficacy can be drawn.
Peptide stability depends strongly on temperature, moisture, and pH. Lyophilized Selank is generally most stable when stored cold and dry, with freezer temperatures commonly used for long-term storage. In solution, the compound is susceptible to hydrolysis and to microbial growth if it is not handled aseptically. The C-terminal proline-rich extension appears to slow enzymatic cleavage relative to tuftsin, though quantitative degradation rates vary with the matrix and the conditions tested. Published stability data specific to Selank remain sparse.
Quality assessment of Selank samples typically combines purity determination with identity confirmation and counter-ion analysis. Purity is usually reported as a percentage by chromatographic area, with values above 95 percent often quoted for research-grade material. Water content and residual solvents are checked in lyophilized batches because they affect both stability and accurate mass determination. A reported purity figure does not by itself establish that a sample is the intended sequence, so orthogonal methods are needed to rule out sequence isomers or truncation products.

Selank is not a naturally occurring peptide and has no known endogenous counterpart in human physiology. Russian-language sources frequently call it TP-7, while English-language sources use the name Selank almost exclusively. Database indexing is uneven, partly because early reports appeared in regional journals that are not widely cataloged. Some summaries describe the material as a tuftsin analog and others as a synthetic heptapeptide; the labels overlap rather than conflict. Citing the primary sequence resolves ambiguity more reliably than the research or trade name alone.
Selank is a synthetic heptapeptide with the sequence Thr-Lys-Pro-Arg-Pro-Gly-Pro, written TKPRPGP in one-letter notation. Its structure consists of the immunomodulatory tetrapeptide tuftsin, Thr-Lys-Pro-Arg, extended at the carboxyl terminus by a Pro-Gly-Pro segment. The molecular formula is commonly given as C33H57N11O9, corresponding to a monoisotopic mass near 751.4 Da and an average molecular mass near 751.9 Da. All seven residues are proteinogenic amino acids, and the molecule carries no modified side chains or non-natural linkages.
The compound was designed at the Institute of Molecular Genetics of the Russian Academy of Sciences during the 1980s and 1990s. The stated design goal was to retain the immunomodulatory and central nervous system activity attributed to tuftsin while improving resistance to enzymatic breakdown. Adding a proline-rich tail to the short parent peptide was a deliberate strategy, because proline residues restrict the conformations available to many peptidases. The same laboratory produced Semax, an ACTH fragment analog, and both compounds were developed in parallel as short, enzymatically stabilized peptides intended for intranasal use.
Animal studies have examined behaviour in tests of anxiety, memory retention and stress response, and several report changes in neurotrophic or neurotransmitter-related markers. The human evidence base is much smaller, consisting mainly of short trials conducted in Russia with limited reporting in English-language journals. Sample sizes are modest and outcome measures vary between studies, so the findings are best described as preliminary. Independent replication under modern trial standards has not been widely reported.
Outside its country of origin the compound is generally handled as a research chemical rather than an approved medicine. No regulatory approval from the United States Food and Drug Administration or the European Medicines Agency has been granted for human use. Identity and purity are normally checked by reverse-phase high-performance liquid chromatography, with mass spectrometry used to confirm the molecular mass. Lyophilised material is stored cold and desiccated, and repeated freeze-thaw cycles are avoided.
Selank is a synthetic heptapeptide with the sequence Thr-Lys-Pro-Arg-Pro-Gly-Pro. It was designed at the Institute of Molecular Genetics of the Russian Academy of Sciences as a structural analogue of tuftsin, a naturally occurring tetrapeptide fragment of the immunoglobulin heavy chain. The added Pro-Gly-Pro tail was intended to slow enzymatic degradation and extend biological activity. In Russia it is registered as an anxiolytic nasal preparation, while regulators elsewhere have not approved it for clinical use.
Proposed mechanisms centre on modulation of the GABAergic system, with reports of altered expression of genes related to GABA-A receptor subunits and changed monoamine turnover. Some studies describe inhibition of enkephalinase, the enzyme that degrades endogenous enkephalins, which may prolong opioid peptide signalling. Effects on brain-derived neurotrophic factor and on cytokine expression have also been reported. These findings come largely from animal models and small human studies, and the precise primary target remains unresolved.
Claire fixes up his wound and a bond develops between the two; she allows McCormick to stay, and he and Nat later build a simulated bomber-plane cockpit in Nat's treehouse so that McCormick can teach Nat how to fly. McCormick passes out and is hospitalized, where he discovers his body is failing as his age begins to catch up to him. McCormick tracks down Finley's daughter Susan, who informs him her father died in a lab fire before she was born. The FBI discovers there was a warehouse lab fire in the early 40s, and all the scientists including Finley died trying to save the frozen McCormick from the chaos. In the aftermath of the fire, people mistook the cryonics chamber for a water heater, not knowing that McCormick was inside, resulting in him being accidentally frozen for 53 years. She gives McCormick her father's journals, detailing the cryogenic process, and Finley's notes disclose that the rapid aging is irreversible. Susan also reveals that Helen is still alive, but they escape before the FBI, who is now after McCormick, catch up to them. Claire drives McCormick to an air show where he commandeers a B-25 bomber to fly to Helen's seaside-lighthouse home, with Nat stowing away on board. Claire gives Harry's journals to the FBI, for their plans to replicate and modernize the experiment. McCormick suffers another aging attack, forcing Nat (who is now slightly familiar with the plane's controls after his simulated-training session with McCormick) to land the plane in the field near Helen's house.
=== Alcohol-induced dose dumping (AIDD) === It is by definition an unintended rapid release in large amounts of a modified-release dosage due to a co-ingestion with ethanol. Some interactions between alcohol, biological factors and the presentation of the drug can influence the apparition of AIDD by:
=== Scotland to England in 1984 === He began at the age of 18 at John o' Groats (at the northern tip of Scotland) in 1984, and walked to Land's End in England. On this journey, which he carried out with three other people, he pushed a wheelchair 1,000 miles (1,600 km) and raised £3,500 for The Forelands School for handicapped children. In 1983, he had already run 21 miles (34 km) around a 400-metre track to raise further money for The Forelands School for handicapped children, at Broadstairs in Kent.
=== Vestibule === The vestibule is the area between the labia minora, where the vaginal introitus and the urinary meatus - the openings of the vagina and urethra respectively - are located. The meatus is below the clitoris and atop the introitus. The introitus is sometimes partly covered by a membrane called the hymen. The hymen will usually rupture during the first episode of vigorous sex, and the blood produced by this rupture has been seen to signify virginity. However, the hymen may also rupture spontaneously during exercise or be stretched by normal activities such as the use of tampons and menstrual cups, or be so minor as to be unnoticeable, or be absent. In some rare cases, the hymen may completely cover the introitus, requiring a surgical procedure called a hymenotomy. Two greater vestibular glands known as Bartholin's glands open into either side of the introitus and secrete a mucous vaginal lubricant. The openings of the lesser vestibular glands, known as Skene's glands, are found on either side of the urethral meatus.

The judicial branch is headed by four high courts, consisting of the Supreme Court which deals with penal and civil matters, the Council of State, which has special responsibility for administrative law and also provides legal advice to the executive, the Constitutional Court, responsible for assuring the integrity of the Colombian constitution, and the Superior Council of Judicature, responsible for auditing the judicial branch. Colombia operates a system of civil law, which since 1991 has been applied through an adversarial system. Despite a number of controversies, the democratic security policy has ensured that former President Álvaro Uribe remained popular among Colombian people, with his approval rating peaking at 76%, according to a poll in 2009. However, having served two terms, he was constitutionally barred from seeking re-election in 2010. In the run-off elections on 20 June 2010 the former Minister of Defense Juan Manuel Santos won with 69% of the vote against the second most popular candidate, Antanas Mockus. A second round was required since no candidate received over the 50% winning threshold of votes. Santos won re-election with nearly 51% of the vote in second-round elections on 15 June 2014, beating right-wing rival Óscar Iván Zuluaga, who won 45%. In 2018, Iván Duque won in the second round of the election with 54% of the vote, against 42% for his left-wing rival, Gustavo Petro. His term as Colombia's president ran for four years, beginning on 7 August 2018.

Nonsense mutations comprise around 20% of single nucleotide substitutions within protein coding sequences that result in human disease. Nonsense mutation-mediated pathology is often attributed to reduced amounts of full-length protein, because only 5-25% of transcripts possessing nonsense mutations do not undergo nonsense-mediated decay (NMD). Translation of the remaining nonsense-bearing mRNA may generate abbreviated protein variants with toxic effects. Twenty-three different single-point nucleotide substitutions are capable of converting a non-stop codon into a stop-codon, with the mutations CGA
Sampling provides one rational means of verification that a production lot conforms to the requirements of technical specifications. 100% inspection does not guarantee 100% compliance and is too time-consuming and costly. Rather than evaluating all items, a specified sample is taken, inspected or tested, and a decision is made about accepting or rejecting the entire production lot. Sampling plans have known risks: an acceptable quality limit (AQL) and a rejectable quality level, such as lot tolerance percent defective (LTDP), are part of the operating characteristic curve of the sampling plan. These are primarily statistical risks and do not necessarily imply that a defective product is intentionally being made or accepted. Plans can have a known average outgoing quality limit (AOQL). A single sampling plan for attributes is a statistical method by which the lot is accepted or rejected on the basis of one sample. Suppose that we have a lot of sizes M {\displaystyle M} ; a random sample of size N < M {\displaystyle N<M} is selected from the lot; and an acceptance number B
=== Function === Calcium is an essential element needed in large quantities. The Ca2+ ion acts as an electrolyte and is vital to the health of the muscular, circulatory, and digestive systems; is indispensable to the building of bone in the form of hydroxyapatite; and supports synthesis and function of blood cells. For example, it regulates the contraction of muscles, nerve conduction, and the clotting of blood. As a result, intra- and extracellular calcium levels are tightly regulated by the body. Calcium can play this role because the Ca2+ ion forms stable coordination complexes with many organic compounds, especially proteins; it also forms compounds with a wide range of solubilities, enabling the formation of the skeleton.

== Mechanisms of hepatotoxicity == Since the withdrawal in 2000, mechanisms of troglitazone hepatotoxicity have been extensively studied using a variety of in vivo, in vitro, and computational methods. These studies have suggested that hepatotoxicity of troglitazone results from a combination of metabolic and nonmetabolic factors. The nonmetabolic toxicity is a complex function of drug-protein interactions in the liver and biliary system. Initially, the metabolic toxicity was largely associated with reactive metabolite formation from the thiazolidinedione and chromane rings of troglitazone. Moreover, the formation of quinone and o-quinone methide reactive metabolites were proposed to be formed by metabolic oxidation of the hydroxy group (OH group) of the chromane ring. Detailed quantum chemical analysis of the metabolic pathways for troglitazone has shown that quinone reactive metabolite is generated by oxidation of the OH group, but o-quinone methide reactive metabolite is formed by the oxidation of the methyl groups (CH3 groups) ortho to the OH group of the chromane ring. This understanding has been recently used in the design of novel troglitazone derivatives with antiproliferative activity in breast cancer cell lines.
==== European Union ==== The European Union's (EU) Food Supplements Directive of 2002 requires that supplements be demonstrated to be safe, both in dosages and in purity. Only those supplements that have been proven to be safe may be sold in the EU without prescription. As a category of food, food supplements cannot be labeled with drug claims but can bear health claims and nutrition claims. The dietary supplements industry in the United Kingdom (UK), one of the 28 countries in the bloc, strongly opposed the Directive. In addition, a large number of consumers throughout Europe, including over one million in the UK, and various doctors and scientists, had signed petitions by 2005 against what are viewed by the petitioners as unjustified restrictions of consumer choice. In 2004, along with two British trade associations, the Alliance for Natural Health (ANH) had a legal challenge to the Food Supplements Directive referred to the European Court of Justice by the High Court in London. Although the European Court of Justice's Advocate General subsequently said that the bloc's plan to tighten rules on the sale of vitamins and food supplements should be scrapped, he was eventually overruled by the European Court, which decided that the measures in question were necessary and appropriate for the purpose of protecting public health. ANH, however, interpreted the ban as applying only to synthetically produced supplements, and not to vitamins and minerals normally found in or consumed as part of the diet.
At sub-anesthetic doses, ketamine produces a dissociative state, characterised by a sense of detachment from one's physical body and the external world that is known as depersonalization and derealization. At sufficiently high doses, users may experience what is called the "K-hole", a state of dissociation with visual and auditory hallucination. John C. Lilly, Marcia Moore, and D. M. Turner (among others) have written extensively about their own entheogenic and psychonautic experiences with ketamine. Turner died prematurely due to drowning during presumed unsupervised ketamine use. Recreational ketamine use has been implicated in deaths globally, with more than 90 deaths in England and Wales in 2005–2013. They include accidental poisonings, drownings, traffic accidents, and suicides. The majority of deaths were among young people. Several months after being found dead in his hot tub, actor Matthew Perry's October 2023 apparent drowning death was revealed to have been caused by a ketamine overdose, and, while other factors were present, the acute effects of ketamine were ruled to be the primary cause of death. Due to its ability to cause confusion and amnesia, ketamine has been used for date rape.
Hypothalamic–pituitary hormones are hormones that are produced by the hypothalamus and pituitary gland. Although these organs are relatively small, the hormones have effects throughout the body. They can be classified as a hypothalamic–pituitary axis (HP axis), of which the adrenal (HPATooltip hypothalamic–pituitary–adrenal axis), gonadal (HPGTooltip hypothalamic–pituitary–gonadal axis), thyroid (HPTTooltip hypothalamic–pituitary–thyroid axis), somatotropic (HPSTooltip hypothalamic–pituitary–somatotropic axis), and prolactin (HPPTooltip hypothalamic–pituitary–prolactin axis) axes are branches.

Reports describe modulation of GABA signalling, changes in monoamine turnover and effects on neurotrophic factor expression. These observations come mainly from animal and cell studies. A single unifying mechanism has not been demonstrated.
The peptide enters plasma rapidly and is broken down by ordinary proteases into amino acids and shorter fragments. Reported half-lives are short. Whether meaningful amounts of the intact molecule reach the brain is an open question.
Most clinical reports are small, published in Russian and not independently replicated. English-language reviews highlight the absence of large randomised trials. Conclusions about efficacy should be treated as provisional.
The most common approach combines reverse-phase liquid chromatography with mass spectrometry. Chromatography separates the components while mass spectrometry confirms the molecular mass. Peptide sequencing or tandem mass analysis can further verify the amino acid order.
